چکیده

The accurate ab initio quantum chemical (QM) method multiconfigurational second-order perturbation (CASSPT2)/complete active space self-consistent field (CASSCF) has been used in conjunction with molecular mechanics (MM) procedures to compute molecular properties and photoinduced reactivity of DNA/RNA nucleobases (NABs) in isolation and within a realistic environment, in which the double helix strand, the aqueous media, and the external counterions are included. It is illustrated that the use of an MM model is helpful both to account for shortand long-range effects of the system surrounding the QM molecular core and to provide the proper structural constraints that allow more accurate QM geometry determinations.

The role of pyrimidine nucleobase excimers in DNA photophysics and photoreactivity*

Quantum chemical studies using the accurate CASPT2//CASSCF procedure show that π-stacked interactions in biochromophores such as pyrimidine (Pyr) DNA/RNA nucleo bases pairs yield excimer-like situations which behave as precursors of processes like charge transfer (CT) or photoreactivity and are the source of the emissive properties in DNA. Examples are the CT between adjacent DNA nucleobases in...

Nature of Lithium Interactions with DNA Nucleobases: Theoretical Study

In the present study, the interactions of three different lithium species Li+, Li3+ and CH3Li with several different sites of the most stable tautomers of DNA nucleo-bases are presented. This investigation is based on the results of thermochemical properties, Quantum Theory of Atoms in Molecules (QTAIM) and Natural Bond Orbi...

Photochemically Induced Binding of Aflatoxins to DNA and Its Effects on Template Activity1

involvement of this functional group in the toxicological action. It is noteworthy that pobycyclic anomatics are carcinogenic under the influence of various activations (17, 29, 39, 59). It is also possible that the photoreactivity of AFB1 for its potential as a DNA binder simulates the well-established microsomal activation of aflatoxins. Microsomal activation of cycbophos phamides also induce...

[Synthesis and characterization of model system in biological related function--model compounds for investigation of metal-metal interaction and nucleobase-aromatics interaction].

Several kinds of compounds available as model systems to investigation on metal-metal interaction in metalloproteins and on interaction of aromatics with nucleobases in arene intercalation to DNA were synthesized and characterized by spectrometry. Double and triple layered porphyrinophanes linked with 1,1'- or 1,3-disubstituted ferrocene moieties and their iron and cobalt complexes were prepare...

Structural Determinants of Photoreactivity of Triplex Forming Oligonucleotides Conjugated to Psoralens

Triplex-forming oligonucleotides (TFOs) with both DNA and 2'-O-methyl RNA backbones can direct psoralen photoadducts to specific DNA sequences. However, the functional consequences of these differing structures on psoralen photoreactivity are unknown. We designed TFO sequences with DNA and 2'-O-methyl RNA backbones conjugated to psoralen by 2-carbon linkers and examined their ability to bind an...
